Abstract
It has been established that calcium may inhibit intestinal iron absorption. In health centers, combination of 200 mg ferrous sulphate and 500 mg calcium lactate are often administered to pregnant women. The question is whether such administration is effective in controlling iron-deficient anemia that commonly occurs in pregnant women. This study was intended to find the effect of ferrous sulphate and calcium lactate combination administration. The doses used in this study based on human doses commonly applied in health centers and were converted to animal doses. Other doses by doubling ferrous sulphate or calcium lactate of iron status in rats with iron deficiency. This study used 75 female wistar strain rats of 4 week old. The design was factorial 2 x 2 post test only design. Before treatment, rats were rendered to be iron-deficient by giving low iron food for 7 weeks. Rats were then divided into 4 treatment groups and administered with ferrous sulphate and calcium lactate combination as follows: PI = FeSO4 3.6 mg/day and calcium lactate 9 mg/day; P2 = FeSO4 3.6 mg/day and calcium lactate 18 mg/day; P3 = FeSO4 7.2 mg/day and calcium lactate 9 mg/day; P4 = FeSO4 7.2 mg/day and calcium lactate 18 mg/day; for 2 weeks. In this study serum iron (SI) concentration, total iron binding capacity (TIBC), percentage of transferrin saturation, and hemoglobin were used as parameter for iron status. Data were analyzed using one-way ANAVA test and factorial test. Result showed that FeSO4 and calcium lactate combination administration to all treatment groups increased SI, reduced TIBC, and increased transferrin percentage saturation significantly (p<0.05), while hemoglobin concentration showed no increased (p>0.05) compared to that in iron deficiency before treatment. Factorial test revealed that there was no significant (p>0.05) effect of FeSO4 and calcium lactate combination administered on the increase of rat's iron status in all treatment groups.
