Abstract

Diagnosis of tuberculosis is still being established based on direct microscopy examination of the sputum and followed examination culture is more sensitive and specific for confirmation of identification. Solid medium of culture germs are the most accurate ways of checking but has disadvantages in timing growth. In this research, medium culture used were added earthworm (Lumbricus rubellus) flour on the grounds of this type of worm is the most widely cultivated in Indonesia and has amino acids that are higher than meat and fish as well as makronutrient and mikronutrient contains a much needed microorganism to optimate growth.This research is a experiment research of the results of laboratory observations aiming to analyze the difference in Mycobacterium tuberculosis H37Rv ATCC 25177 growth on the nutrient agar medium added earthworm (Lumbricus rubellus) flour compared to the growth on Lowenstein Jensen medium. The result of research was showing that on the average Lowenstein Jensen medium has 11,0 days of colonies growth. There are difference in the old growth of Lowenstein Jensen medium on nutrient agar medium added earthworm (Lumbricus rubellus) flour with 25% consentration has 46,6 days of colonies growth, 50% has 37,4 days, 75% has 27,8 days and 100% has 46,6 days of colonies growth. Statistical analysis showed no significant difference(p=0,478 >α (0,05) in the old growth of Lowenstein Jensen medium on nutrient agar medium added earthworm (Lumbricus rubellus) flour with 25%, 50%, 75% and 100% consentration concluded that nutrient agar medium added earthworm (Lumbricus rubellus) flour can using as an alternative medium for the growth of Mycobacterium tuberculosis H37Rv ATCC 25177 but with growth time longest and less growth coloniest.
