Abstract
Candida albicans have been shown to be resistant to commercial antifungal. Exploration of new antifungal were done to conquer that problem. Biofilm protected Candida albicans from antifungal penetration. Biofilm are different from planktonic cell. β-1,3-glucan content was increased in Candida albicans cell walls and extracellular matrix from a biofilm compared to planktonic cell. β-1,3-glucan was responsible in minimizing antifungal penetration on Candida albicans biofilm. Virgin Coconut Oil (VCO) have antifungal properties to Candida albicans cell culture, but never been tested to Candida albicans biofilm. A common antifungal, nystatin, was used. β-1,3-glucanase enzyme was used in combination with VCO and nystatin to increase the antifungal activity. Antifungal activity were measured quantitatively by viability analysis and weight measurement of Candida albicans biofilm. Viability analysis is based on live cell ability to reduced tetrazolium XTT to form colour formazan. According to viability analysis, VCO 25% (v/v) had inhibition activity to Candida albicans biofilm growth for 44,4%. Nystatin 80 units/ml inhibited Candida albicans biofilm growth to 36,3%. While combination of VCO and β-1,3-glucanase elevated VCO activity to 61,6%, affiliation of nystatin with β-1,3-glucanase increased nystatin activity until 26,7%. This results showed that β-3-glucanase can elevated VCO or nystatin activity on Candida albicans biofilm growth. Weight measurement of Candida albicans biofilm showed different pattern of antifungal activity compared to viability analysis. Since weight measurement did not correlate directly with cell growth in biofilm, antifungal activity were counted based on viability analysis.
