Abstract

This study has identified six phenolic compounds in methanol extract of the flesh of kepel fruit (Stelechocarpus burahol), two of which were successfully obtained as isolates. Methanol extract of the flesh of kepel fruit was obtained using the maceration method. Fter filtration, the filtrate was collected and concentrated to dryness under reduced pressure in a rotary evaporator to yield methanol extract and 18,76 % rendement was obtained. The phenolic compounds contained in methanol extract of the flesh of kepel fruit were analyzed under the Follin Ciocalteau method, resulting in 38,23 mg/L. Antioxidant activities tests of methanol extract of the flesh of kepel fruit were done using two methods, 2,4- DPPH radical scavenging assay and lipid peroxidation using ferric thiocyanate test. Both methods resulted that the methanol extract of the flesh of Kepel fruit has antioxidant activity. From the DPPH assay, IC50 value of methanol extract of the flesh of kepel was 441,64 ppm. Separation of the chemical components in methanol extract of the flesh of kepel fruit was done using various chromatographic methods to obtain six major spots and two of which could be obtained as isolates. Analysis of the six spots using TLC scanner with UV-Vis detector produces the maximum absorption peak at #955;max= 270-310 nm which is the maximum absorption peak area/range of the phenolic compound group. Analysis of the 1H-NMR and 13C-NMR spectra of the two isolates were incapable to provide clear information since both isolates still had lipid content as contaminant. However, signals in the aromatic region were observed as para substituted benzene ring with a hydroxyl group unit, so these two isolates were suggested as phenolic compounds.
