Abstract

The aim of this study is to optimize the co-cultivation period with RNAi gene PFP transformation in sugarcane (Saccharum officinarum) mediated by A. tumefaciens. the research is used Agrobacterium tumefaciens strain GV 3101. Callus of three sugarcane varieties are varieties Bululawang (BL), PS 862 and HW is used as explants transformation. Induction of callus of sugarcane leaf roll performed in vitro culture. Transformation MS medium supplemented with 3 mg /L 2,4-D and 0.5 mg / L kinetin Callus age of 8 weeks (BL and PS 862 varieties) and 12 weeks (HW) is used as explants for. Co-cultivation period treatment 24 hours and 72 hours was conducted on solid MS medium with 100 μM asetosiringone. Concentration of 100 mg/L kanamycin was obtained as the best concentration in the selection medium used to distinguish between the transformant and nontransforman. The results showed that the efficiency of transformation in the 72 hours co-cultivation period give higher transformation efficiency than 24 hours co-cultivation period. Transformation efficiency with 72 hours co-cultivation period on Bululawang , PS 862 , and HW sugarcane varieties are 13 % , 5.5 % and 0 % . While, the transformation efficiency with 24 hours co-cultivation period on Bululawang , PS 862 and HW sugarcane varieties are 0 % , 0.6 % and 0 % . Transformation experiments resulted in 39 putative transgenic plants in sugarcane varieties Bululawang and 10 putative transgenic plants at PS 862 varieties .The presence of RNAi gene pfp (1,110 bp) was confirmed by PCR, and is founded in a Bululawang planlet and a PS 862 planlet .
