Abstract

Bitemark analysis has been done by looking at the peculiarities and characteristics of tooth formation of physical evidence that is then compared with the teeth of suspects to find the person who left a trace of the bitemark. Bitemark analysis is difficult to solve because it often is subjective and depends on the experience and the procedures used by odontologist. Hence, now it begins to be considered that saliva deposited during biting can be used as an alternative way of bitemark analysis, primarily because saliva contains DNA. This study aims to determine the isolation of human DNA from a bitemark on Malang apple (Paynes malus) which can be used as a forensic identification material. The sample is human DNA contained in a bitemark at the Malang apple (Paynes malus) and blood as a positive control, with a sample of 3 apples. Deposited saliva on the bitemark is taken with the double swab technique. DNA isolation is performed by using the Chelex method. After the isolation process, measurement of DNA content and purity is performed. Of the three samples, it is obtained two samples that meet the requirements to do the typing (levels> 20μg / ml and purity ranged between 1-2), the samples of apple 1 and 2. The two samples along with blood samples were performed PCR amplification using locus CSF1PO, THO1, TPOX, and VWA. PCR amplification was not performed on the sample of apple 3 because the value of purity is less than 1. Visualization of electrophoresis showed that the DNA sample was detected by the four locus. It can be seen from the presence of DNA band in the samples of apple 1 and apple 2 for each locus and the band pattern of the two samples are in line with the band of blood samples. Thus, isolation of human DNA from the bitemark on the malang apple (Paynes malus) can be used as a forensic identification material.
