Abstract

Temozolomide is an imidazotetrazine derivative of the alkylating agent with chemical structure, unique pharmacokinetic. Temozolamide is imidazotetrazine genetrasi prodrugs that undergo both spontaneous conversion to the active lorm MTIC which does not require metabolism in the liver. Temozolomide has a small molecular weighl, so that efficient Temozolomide crosses the blood-brain barrier. Temozolomide lncreased survival and an increase in time to progression (TTP). This was without having a statistically significant negative effect on quality of life and with a low incidence of early adverse events. Crade 3/4 haematological toxicity was found in 5 to 14%. ln recurrent high grade gliomas, temozolomide did not increase overall survival but increase time to progression.
