Abstract

Cervical cancer is the second most common cancer in women in the world, and the most frequent cancer in Indonesia. The cause of the development of cervical cancer is infection of Human Pappilomavirus (HPV). HPV types that cause invasive cervical cancer in the world are HPV 16, 18, 33, 45, 31 and in Asia are HPV 16, 18, 58, 33, 52. HPV types that cause cervical cancer are HPV 16 and 18. HPV types at Jakarta, Tasikmalaya, and Bali are HPV 52, 16, 18, and 39 (Vet JNI, et al, 2008), in Jakarta, HPV 16, 18, and 52, (Schellenkens MC, et al, 2004), and Bandung, namely HPV 16, 18, 45, da 52 (Panigoro R, et al, 2013; Tobias MDL, et al, 2014). Primary prevention of cervical cancer is the HPV vaccination (Andrijono, 2007). Currently there are two types of HPV prophylactic vaccines are high risk Vasin fatherly bivalent HPV 16 and 18 (Cervarix) and high-risk HPV types and low risk quadrivalen well as the vaccine for HPV 16, 18, 6, and 11 (Gardasil). Primary prevention can be done by knowing the types of HPV that infect patients with cervical cancer in Indonesia first. Data types of HPV in Indonesia is still limited and does not represent the whole of Indonesia. Data types of HPV that infect patients with cervical cancer in Surabaya is still no report. Research was conducting at Dep Anatomical Patology Dr Soetomo Hospital Surabaya, using 53 paraffin blocks of patients CIN I, CIN II, CIN III, SCC derived from patients aged between 26-79 years. CIN I and CIN II including LSIL group, whereas CIN III include HSIL. Paraffin blocks were cut with a microtome collected and used for DNA extraction. DNA extraction product used for PCR with the product of about 202bp to control and 139-145bp to sample. HPV genotyping is done by using Ampliquality HPV-Type express (AB analitica) with reverse line blot technique that can be used to detect 40 types of HPV that includes a group of high risk and low risk. HPV types that can be detected, namely 6, 11, 16, 18, 26, 31, 33, 35, 39, 40, 42, 43, 44, 45, 51, 52, 53, 54, 55, 56, 58, 59, 61, 62, 64, 66, 67, 68 (AEB), 69, 70, 71, 72, 73, 81, 82, 83, 84, 87, 89, 90. Resulf of this research shown that HPV infected cervical LSIL were HPV 16 and 18. HPV infected cervical HSIL were HPV 16 and 52. HPV infected cervical SCC were HPV 16 , HPV 18, HPV 52, HPV 45, HPV 16/18, HPV 16/45. Prevalence of HPV single infection in pre cancer lession and SCC cervix were 92,45% (49/53) which HPV 16, HPV 18, HPV 45, HPV 52, and HPV 67, and multiple infection were 7,55% (4/53) which HPV /18, HPV 16/45, HPV 16/67, HPV 52/67. Prevalence of HPV genotype infected pre cancer lession and SCC cervix were HPV 16 (75,5%), HPV 18 (15,1%), HPV 52 (7,5%), HPV 67 (5,7%), and HPV 45 (3,8%). Conclution, the most prevalence HPV genotype in the pre cancer lession and SCC cervix are HPV 16, 18, 52, 67, and 45. HPV 52, 67, and 45 should be considered as the vaccine type in the HPV second vaccine generation.
