Abstract

Background: Chitosan is a natural polymer with favourable properties such as biocompatibility, biodegradability, non- allergenic, and non- toxic which make it a very promising material for scaffold in tissue engineering. Application of chitosan based systems in hard tissue engineering such as bone, cartilage, and periodontal are reviewd with recent and relevant examples. Hydroxiapatite is a polymer of calcium phosphates from bones and can be extracted cortical bovine bone. Hydroxiapatite has poor solubility, so it is combined with chitosan because chitosan has high solubility to make good strength scaffold for bone healing.. Purpose: The purpose of this laboratory research is to study the density of collagen on bone with the application of hydroxyapatit chitosan and hydroxyapatite. Methods:. The samples used were 24 healthy 3 months old male wistar’ s rats, with approximately 200 grams of body weight and they were devided into 4 groups. Two groups were taken for hydroxiapatite chitosan and the others for hydroxiapatite. A group were taken for hydroxiapatite chitosan was given for 7 days in group C7, C14 for 14 days. A group were taken for hydroxyapatite was given for 7 days in group H7, H14 for 14 days. Seven days of operation time was waiting before the sample being decapited for further histological examination in the post operation. A hematoxillin staining was used and the density of collagen counted under light microscopy with 400 times magnification T- test and least significant difference test were used for the statistic evaluation. Result:. shown increase of the density of collagen in hydroxiapatite chitosan group of 7 days and the hydroxiapatite chitosan of 14 days is decrease. On the other hand, the result of study shown decrease of the density of collagen in hydroxiapatite group of 14 days and it increase for the group of 7 days. This study that duration time of hydroxiapatite and chitosan had not been interfered significantly with the density of collagen during bone healing after the defect in wistar’s rats. Conclusion: There is no different between hydroxyapatite and hydroxyapatite chitosan application to the density of collagen. Chitosan only can accelerate the bone healing in proliferation phase.
