Abstract

Background: Stroke is one of the most devastating diseases and a leading cause of death and disability in the world with further impact to emotional and economical problems. This research mainly to investigate the role of intraventricular transplantation using bone marrow mesenchymal stem cell in stroke. Material and Methods: This study was a one group (eight patient) pre and post test design. Subjects were randomized from supratentorial hemorrhagic stroke patients, after six months treatment with stable neurological deficits. One control group (eight patient) were included to compare the clinical outcomes. Clinical outcomes were measured using NIHSS scale three months after treatment. Meanwhile expression of neurogenesis biomarkers (BDNF, VEGF and Vimentin) were assessed on subject’s cerebrospinal fluid on pre and one-month post treatment. Results: The result showed significant correlation between the median score value of NIHSS before and after treatment. No correlation between the level of BDNF (p= 0,422), VEGF (p= 0,260) and Vimentin (p= 0,256) pre and post treatment was found. There were evidences of correlation between level of BDNF (p= 0,024; r= 0,773) and VEGF (p= 0,003; r= 0,892) in cerebrospinal fluid based on groups of age. Conclusion: Intraventricular transplantation of BM-MSCs is safe and improved functional status in post-hemorrhagic stroke patient. There were no increased expression on neurogenesis biomarkers (BDNF, VEGF and Vimentin) despite the improvement of functional status, other cellular mechanism could be responsible.
