Abstract
The case of preterm labor becomes one of the health problems in the world because it can cause pain and perinatal death. One of the medical efforts that can be done in preterm labour is a tocolytic agent therapy. In Indonesia, the most widely used tocolytic agents are isoxsuprine and nifedipine. The presence of complaints about side effects during and after use of these drugs led to need for scientific studies related to the effectiveness and safety of nifedipin and isoxsuprine. This study was conducted to analyze effectiveness and safety both of these drugs with parameters : delivery delay, blood preasure and adverse events reactions (ADRs). This study was an observational study conducted by consecutive sampling in pregnant women with a diagnosis of prematurus imminens. Data was collected from 05 April to 05 July 2016 at General Hospital-Prof Dr WZ. Johannes. This study had received approval of the Ethics of Faculty of Medicine, University of Nusa Cendana Kupang. The inclusion criteria were pregnant women with 28-36 weeks gestation age (N = 17). The isoxsuprine was given 10 mg infused intravenously along with 500 mL ringer lactate with a drip rate of 20 drops per minute and for nifedipine group received an initial dose of 20 mg followed by 10 mg every 8 hours until the contraction was lost. Succesfull tocolysis was achived in 86,67% of isoxuprine treated group and 100% of nifedipine treated group. Side effects such as pulmonary oedema, spasm, tachycardia and rashes developed in isoxuprin and complaint of dizziness was occurred in nifedipine. The results of measurement of blood pressure of patients before and after therapy showed a change in systolic and diastolic blood pressure in the range 90-120 mmHg / 50-80 mmHg, so that required monitoring of changes in blood pressure of patients during therapy.
