Abstract

This study aims to determine chromatogram profile of rhizomes extract of Kaempferia rotunda L. as an alternative to the quality control of herbal medicine by KLT-Dencitometry method. Rhizomes were collected from Materia Medika Batu (standart), Kebun Raya Purwodadi Pasuruan and market of Genteng Surabaya and extracted with etanol 70% using maseration method. The chromatogram profile of both extracts were made by thin layer chromatography using silica gel 60F 254 as stationary phase and toluen – ethyl acetat (85:15) as development solvent. Spotting was done using 4 μl volume by Linomat V and eluted within Chamber. Camag TLC Scanner 4 were used for peak analysis and scanned at wavelenght 254 nm and 366 nm. Spectra analysis at wavelenght 200 nm – 400 nm. The selection of characteristic peaks based on several criteria such as precise, pure and selective. The results showed that peaks with Rf of 0.14 and 0.24 were selected as the characteristic peaks of rhizomes extract of Kaempferia rotunda L. For identification of rhizomes extract as raw material of herbal medicine, then both peaks must arise with criteria of Rf value, peak identity and peak purity that meet the requirements.
