Abstract

The purpose of this in vitro research was to determine the effect of the dosage and the length of application time of pulperyl towards the death offibroblas cell. The sample used in this research was BHK-21 cell taken from the fibroblas cell of hamster 's kindney implanted in microplate 96 wells. The pulperyl doses given in this research were 0.02, 0.03, 0.04, 0.5, 0.06, 0.07, and 0.08 cc while the duration ofpulperyl applied was 24. 48 and 72 hours. The sample was divided into the dose and the duration of the pulperyl applied that there would be 24 group of samples which each group had 8 wells. The numbers of BHK-21 cells alive could be seen using MIT (3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) enzymatic essay tests. The numbers ofdeadfibroblas cells were counted from the decreased of alive fibroblas cells percentage. The analyses result statistically showed that the doses and the duration ofpulperyl applied affected the number or the percentage of the alive fibroblas cells (p < 0.05). Generally, it was compared to the 0.02 cc dose of pulperyl; started from 0.05 cc dose, the number of alive fibroblas cells decreased lower significantly (p < 0.05), while the duration of pulperyl applied for 24 hours also had significantly higher towards the duration of the pulperyl applied for 48 and 72 hours (p < 0, 05). In addition to that, it was found that there was significant relationship (p < 0.05) between the percentage of alive fibroblas cells and the doses ofpulperyl and the duration ofpulperyl applied where the higher the dose ofpulperyl and the longerpulperyl applied, the higher the percentage of dead filbroblas cells. It can be concluded from this research that the dose and the duration ofpulperyl applied influenced the decreased of the percentage of alive fibroblas cells. The higher the dose and the longer the duration of pulperyl applied, the higher the percentage of the dead fibroblas cells.
