Abstract

Andrographolide has low bioavailability due to its low water solubility and short half-life (t1/2 ± 2 hours). Andrographolide-chitosan microparticle was expected to improve bioavailability of andrographolide. Andrographolide-chitosan microparticle was prepared by ionic gelation method-freeze drying with different concentration of chitosan using tripolyphosphate (TPP) as the cross linker. The objective of this research was to investigate the effect of chitosan concentration on physical characteristics such as organoleptic, yield, morphology, infrared spectrum, DTA thermogram, X-ray diffraction analysis, redispersion test, and entrapment efficiency of Andrographolide-chitosan microparticles. Microparticles formed as white powder, odorless, slightly bitter, and easy to redisperse in aquadest. The formulas had spherical shape and smooth surface. The DTA thermogram and X-ray diffraction analysis indicate that the Andrographolide has been trapped in the microparticle hence the crystalinity reduced. Infrared spectrum indicates that there was ionic bonding between chitosan and TPP. Based on entrapment efficiency of andrographolide in the microparticle showed that increasing amount of chitosan decrease andrographolide entrapment in microparticle.
