Abstract

Diarrhea is one of the health problems in Indonesia. One of the factors that can cause diarrhea is the infection of human pathogenic bacteria. One of the human pathogenic bacteria is Shigella flexneri. Shigella flexneri can cause bloody diarrhea with mucus in human. People in the region of North Kolaka of Southeast Sulawesi Province commonly use the extract of torch ginger (Etlingera elatior) seed as the alternative medicine to cure the abdominal pain and diarrhea. This research goal is to determine the antibacterial activity of Etlingera elatior seed ethanol extract against Shigella flexneri in vitro by diffusion method. In this research, diffusion method was applied on 0,5 McFarland Shigella flexneri by using Mueller Hinton agar plate given the ethanol extract solution of torch ginger seed in serial concenteration of 0.125 g/ml, 0.25 g/ml, 0.5 g/ml, 1 g/ml, and 2 g/ml. The ethanol extract of torch ginger was solved in methanol 96% solution. Ciprofloxacin 5 mcg was used as the positive control and the solvent is used as negative control. The result was influenced by the concenteration level of torch ginger seed ethanol extract. By the concenteration of 2 g/ml the average diameter of inhibition zone is 17.8 mm. By the concenteration of 1 g/ml the average diameter of inhibition zone is 14 mm. By the concenteration of 0.5 g/ml the average diameter of inhibition zone is 11. The solution with concenterarion of 0.25 g/ml and 0.125 g/ml does not produce the inhibition zone. The diameter of inhibition zone by the positive control is still bigger than the diameter of zone inhibition produced by the highest concenteration of ethanol extract of torch ginger seed (2 g/ml). Negative control does not produce inhibition zone against Shigella flexneri. From the result it can be concluded that the ethanol extract of torch ginger seed can inhibit the growth of Shigella flexneri in concenteration of 0,5 g/ml.
