Abstract

Background: HHV-6 and HHV-8 have enhanced pathogenicity in the immunocompromised host. Human herpesvirus-6 (HHV-6) and human immunodeficiency virus (HIV) are both tropic for CD4+ lymphocytes. Human herpesvirus 8 infection in patients with HIV/AIDS can caused Kaposi's sarcoma. Early detection of the presence of HHV-8 can prevent the onset of clinical manifestations appear. Aims: In Indonesia, there has been no examination of HHV-6 and HHV-8 infection in HIV-positive patients. This research aims to detect anti-HHV-6 IgG antibodies. It is used to predict an active infection and antigen HHV-8, and the relationship of CD4+ count with anti-HHV-6 antibodies and HHV-8 antigens. Material and methods: Serum samples from 76 HIVpositive were taken from patients that check-up and treatment at hospital or care center in Surabaya. Serum samples were tested for the presence of anti-HHV-6 IgG antibodies using a commercial ELISA kit (Abnova, Cambridge, UK) by indirect ELISA method and HHV-8 (Abbexa, Cambridge, UK) by sandwich ELISA method. Results: anti-HHV-6 IgG antibodies detected in 86.8% percent from total samples. High titer of anti-HHV-6 IgG antibodies detected 24%, medium 47% and low 14%. The presence of HHV-8 infection in HIV-positive did not differ either between males and females or among different age groups. CD4+ cell count showed no association with anti-HHV-6 IgG antibodies. HHV-8 antigen was assessed in 76 HIV-positive samples, and there are 9/76 (11.8%) positive HHV-8 antigens. Patients with HHV-8 antigen have not shown any skin lesions of Kaposi's sarcoma. There is asymptomatic infection of HHV-8. Conclusions: anti-HHV-6 IgG antibodies detected 86.8% in samples. HHV-8 antigen was detected in 11.8% of patients HIV positive but it has not shown any Kaposi's sarcoma lesions. Among anti-HHV-6 IgG antibodies and HHV-8 antigens no association with CD4+ count.
