Abstract

Human Gingival Fibroblast (HGF) is the major components of tissue formation in periodontal tissue and periodontal disease may cause the destruction. Regeneration of HGF by accelerating the proliferation requires tissue engineering therapy. Collagen scaffold derived from freshwater fish scales is one of the tissue engineering techniques. Previous studies reveal the similarity characteristic of collagen scaffold derived from Tilapia fish scales (Oreochromis mossambicus) and Gouramy fish (Osphronemus goramy). They have similar biological structures, habitat, equal percentage of collagen content, and the same amino acid arrangement that can increase the proliferation of fibroblasts. Characteristic equations and abundant amounts in Indonesia make Gouramy fish scales as an alternative to tissue engineering materials. In this preliminary research the study focus on the test of viability of HGF cells treated by Gouramy scales collagen suspension in 24 hours incubation. Objective: To study the viability of Humam Gingival Fibroblast (HGF) treated by Gouramy scales collagen suspension in 24 hours incubation. Methods: HGF is taken from healthy gingiva and seeded in 96 well plates. Gouramy scales with concentration of 1000 ppm, 500 ppm, 250 ppm, 125 ppm, 62,5 ppm, 31,25 ppm, and 15,625 ppm were added into each well and incubated for 24 hours. MTT Assay performed to observe the viability of fibroblast cells. Result: The viability of HGF were increased after the addition of Gouramy scales scaffold concentration 15,625 ppm to 250. However the viability was decreased after the addition of gouramy scales scaffold concentration 500 PPM and 1000 concentration. The highest viability of the cells was shown after the addition of 250 ppm. Conclusion: The highest viability of the cells were shown after the addition of 250 ppm and collagen derived from fish scales Gouramy may have potential in tissue engeeneering
