Abstract

Aimed of this study was to determine the presence of Vibrio cholerae in cockles (Anadara granosa) from different wet markets in Surabaya Central. A total of 100 samples from 7 different markets (Pasar Kembang, Pasar Asem, Pasar Gersikan, Pasar Indrakila, Pasar Kedungdoro, Pasar Keputran, and Pasar Pacar Keling) were examined for the presence of Vibrio cholerae. This identification method was done the enrichment using Alkaline Peptone Water, then cultivated on the Thiosulfate Citrate Bile Sucrose agar media to get a specific colony including round shaped, bright yellow, and 2-3 mm in size. Then the colony was tested in Nutrient Agar which already mixed with sodium chloride (NaCl) as much as 0%, 6%, and 8%. the colony that growth in 0% (+) ; 6% (+/-) ; 8% (-) then was proceeded for Oxidation test. 47% of samples were positively contaminated Vibrio cholerae confirmed by the biochemically test and serogroup O1 detection was done by running into PCR procedure. The results showing a negative result and primer dimer were acquired.
