Abstract

The purpose of this research to know cross reaction Paramphistomum cervi protein with serum-Fasciola gigantica by using Western Blot technique. The result can be used as a Paramphistomiasis serologic diagnostic. This research was conducted using Paramphistomum cervi worm collected from the rumen and reticulum of large ruminant especially cattle and Fasciola gigantica worms collected from bile duct and gall bladder of cattle. The worms were crushed and added with Phospate Buffer Saline (PBS) to make homogenates and then centrifuged at a speed 3500 rpm for 15 minutes. Homogenate Fasciola gigantica was immunized to mice to obtain serum anti-Fasciola gigantica at a dose of 200 µg/mice and booster 3 times with a span of two weeks. The homogenates was performed by SDS-PAGE technique to analyze the protein by using Commasie briliant blue staining. Cross reaction of Paramphistomum cervi protein with serum anti-Fasciola gigantica using by Western blot technique and obtained 11 protein bands are 179, 103, 65, 54, 47, 33, 31, 28, 21, 16, 11 kDa.
