Abstract

The aim of this research was to determine the effectiveness of sappan wood (Caesalpinia sappan L.) extract topically can accelerate the density of collagen on incision wound healing in rats (Rattus norvegicus). Twenty male rats were randomly devided into five groups, there were negative control (K-) treated with ointment base, positive control (K+) treated with povidone iodine 10% ointment, treated groups (P1, P2, P3) treated with sappan wood extract ointment 6,5%, 15%, and 30% . Treatment had been given directly on the subcutan incision area topically once a day for fourteen days. The result data of histopatological appereance were processed with Kruskal-Wallis using SPSS. The result of nonparametric test Kruskal-Wallis is 0,044 show that significantly different (p<0,05), and continues to Mann-Whitney U test. The collagen density of incision wound on group P1 significantly different with group P2, P3, K-, and K+, and there are no more significantly different groups. The compounds of sappan wood extract is flavonoid, saponin, tanin, and alkaloid. Flavonoid and alkaloid have ability the ability as an antioxidant that can reduce free radicals. Saponin can stimulated the migration of keratinocytes which are important for reepithalialization process and can stimulate fibroblast for collagen synthesis to the wound area. Tanin serves as an astringent which can cause skin pores shrink, and to stop the exudates to cover the wound area. Research shows that increasing mean of collagen density is on sappan wood extract ointment treatment are group P1 which indicates that is the optimum dose.
