Abstract

Repeated treatment of morphine produces several change in brain and behavior that far outlast their initial neuropharmacological effects. The nature of persistent drug-induced neural plasticity is of interest because it is thought to contribute to the development of drug dependency and addiction. To determine if chronic morphine treatment alters the morphology of prefrontal cortex (PFC) and nucleus accumbens (NAc), this study was designed to evaluate the effects of a selective kappa opioid receptor agonist U-50,488H on the morphological change of PFC and NAc neurons, we have used chronic morphine exsposure to leads dependency of mice. Mice were treated morphine with an escalating doses from 10 mg/kg to 140 mg/kg. Mice receive morphine twice a day daily for a week. At 8th of the day, mice were treated naloxone to precipitated withdrawal syndrome. U-50.488H dose 0.1; 1 and 3 mg/kg were administrated thirty minutes before injection of morphine. This observation shown that the increasing of locomotion reduced significantly by repeated administration of U-50,488H (F(4,49)) = 18.30 p = 0,001) and a significant reduction on the decreasing of weight (F(4, 49) = 3.360 p=0.020). Furthermore, we observed that treatment of morphine produces the morphological change ofPFC and NAc neurons, that was stained by hematoxylin-eosin. The changes are shape, size, density of neuron and apoptosis cell. Compared with control mice, apoptosis cell of morphine-treated animals.
