Abstract

IN VITRO CITOTOXIC EFFECT OF THE MAHKOTA DEWA (PHALERIA MACROCARPA) FRUIT’S EXTRACT ON MYELOMA CELL CULTURE Cancer is still a fearful disease in the world and an ideal medicine has not been found yet. The fruits of mahkota dewa (Phaleria macrocarpa) have been reported to be used as an anticancer agent, however no scientific reports have been published so far. This study is aimed to test the cytotoxic effect of the mahkota dewa fruit’s on cell culture of myeloma in vitro. The mahkota dewa fruit’s were extracted with metanol. The extract were tested for its anticancer activity using viability test method on cell culture of myeloma. The myeloma cell were incubated in RPMI media contained 10% OF Fetal Bovine Serum (FBS). The concentration of extract mahkota dewa fruit’t were 5; 2,5; 1,25; 0,625 and 0,312 mg/ml. The metanol was used as solution control in five different concentration (10; 5; 2,5; 1,25 and 0,625, respectively) while RPMI was used as negative control. Their solution were added into myeloma cell culture in microwell plate and then incubated for 24 hours, at 37ºC in CO2 incubator. Determination of anticancer activity used trypan blue exclusion method, with parameter value was viability of myeloma cell. Analysis of varian of the data showed that extract mahkota dewa (Phaleria macrocarpa) fruit’s have anticancer activity against cell culture of mice myeloma.
