Abstract
Aim: To investigate the influence of vanadyl sulfate on telomerase activities in pancreatic β cells proliferation of streptozotocin-induced diabetic mice.Methods: 25 mice were divided into five groups i.e. placebo, diabetic and treatment groups with three doses of vanadyl sulphate (5, 30 and 100 mg/kg BW).Diabetic mice model was induced twice by intraperitoneal administration of streptozotocin; 100 mg/kg on day 1 followed by 50 mg/kg on day 14.Administration of vanadyl sulphate in three doses was given on day 21 to day 28.The pancreas tissues was taken on day 28; stained with aldehide fuchsin for morphological quantitative analysis also immunohistochemical examination to observe the existence of telomerase activity. Results: Diabetic condition occurred on day 21 after streptozotocin injection and glucose levels increased significantly (p=0.015). In pancreatic islets of the diabetic group, damaged islets, decline in islet size, and degenerative effects on pancreas β cells were observed in comparison to the normal group. Administration of three different doses of vanadyl sulfate for 7 days significantly reduces blood glucose concentration (p<0.001). After treatment with vanadyl sulfate, pancreatic islets of Langerhans in the diabetic group showed a higher number of prominent β cells, intact islets and an increase in islet size compared to the diabetic group. The proliferative effect in diabetic mice treated with vanadyl sulfate showed a significant increase. This is shown by an increase in diameter of the islets (p<0.001) and the number of β cells on a single islet (p<0.001). Immunohistochemical examination shows that vanadyl sulphate is able to stimulate activation of telomerase. The telomerase activation causes a proliferative effect on pancreas β cells and is shown by islets morphology resembling close proximity to normal mice islet morphology visually. Conclusion: According to the histochemical and immunohisto -chemical results, it was concluded that vanadyl sulfate can significantly decrease blood glucose concentration and stimulate activation of telomerases that enhance β cell proliferation in experimental diabetes mice compared to normal mice.
