Abstract

Melia azedarach L. is one of Indonesian herbal medicine that currently was proven to be active as anticancer. In order to develop as standardized herbal medicine, acute and subchronic toxicity of ethyl acetate fraction of Melia azedarach leaves were carried out to fulfill qualification of phytopharmaceutical products registration. For acute toxicity test, a single dose of 5000, 2000, 300, 50, and 5 mg/kg BW of EA fraction of MA leaves were given orally to male mice. The death and toxicity symptoms were observed every 24 hours for 14 days. For subchronic toxicity test, as much as 140, 280, and 560 mg/kg BW of EA fraction of MA leaves were given orally everyday for 28 days. The clinical parameters such as hematology, clinical chemistry and relative organ weight were observed after 28 days. The research showed there was no death and toxicity symptoms found to all mice. Thus, it can be determined that LD50 of the fraction is more than 5000 mg/kg BW, which is practically nontoxic. On subchronic toxicity test, after clinical symptoms observed such as hematology, clinical chemistry and relative organ weight was done and statistically analyzed, there were no toxic effect showed on male and female rats.
