Abstract

The present study was design to determine characteristic and penetration of diclofenac sodium in niosomal system which had been preparated in hidroxypropylmethyl cellulosa 4000 gel. The compositions of niosomal system is diclofenac sodium, Span 20, and cholesterol 1:6:6. There were two formulas in this study. The first formula (formula I) is diclofenac sodium in hidroxypropylmethyl cellulosa 4000 gel without niosomal system and the second formula (formula II) is diclofenac sodium in niosomal system that was preparated in hidroxypropylmethyl cellulosa 4000 gel. The result showed that niosomal system had effect on color and odor, because of the composition of niosomal system. Niosomal system also increased pH and spread diameter of zero load of diclofenac sodium gel. Data analysis showed that pH of formula I is 6,30±0,04 and formula II is 6,94±0,03. Spread diameter of zero load for formula I is 6,17±0,06 and formula II is 7,60±0,20. Abdominal skin from male wistar rat was used as penetration membrane for this study. The penetration rate of diclofenac sodium from formula I is 1,4036±0,0696 μg/cm2/min and formula II is and 1,1180±0,1111 μg/cm2/min. Membrane permeability for formula I is 1,3387.10-4±0,0677.10-4 cm/min and formula II is 1,0847.10-4±0,1069.10-4 cm/menit. The results were analyzed statistically using independent sample t-test with degree of freedom 4 (df) and degree of confident 95%. Research result revealed that the penetration rate and membrane permeability of diclofenac sodium in hidroxypropylmethyl cellulosa 4000 gel with niosomal system are slower than diclofenac sodium in hidroxypropylmethyl cellulosa 4000 gel without niosomal system.
