Abstract

Orally disintegrating tablet (ODT) are gaining attractiveness over conventional tablets especially for patients having difficulty in swallowing such as pediatric, geriatric, bedridden and disable. However, the requirements of formulating these dosage forms with mechanical strength sufficient to withstand the rigors of handling and capable of disintegrating within a few seconds on contact with saliva are inextricable. Paracetamol, an antipyretic medication, was used as a model drug in the formulated ODT. Therefore, the aim of this study was to determine the effect of of gelatin 1% and 2% as a binding agent on the physical characteristics (hardness, friablitiy, disintegration time) and dissolution of paracetamol ODT made by freeze drying. Tablet were evaluated for hardness, friability, disintegration time, and dissolution. The results showed that mechanical strength of the tablet increased by increasing of gelatin concentration but there was no significant difference in dissolution result . The hardness of tablet prepared using gelatin 2% was significantly improved compared to tablet prepared using gelatin 1%. The friability of tablet prepared using gelatin 1% and 2% were less than 1%. All of the formula showed disintegration time less than 60 seconds and percent dissolved within 30 minutes more than 80%.
