Abstract

Ezetimibe has been classified as Biopharmaceutics Classification System (BCS) class II category which is practically insoluble in water and has poor bioavailability. This problem will cause low dissolution rate, absorption and bioavailability. Solid dispersion system could enhance dissolution rate of ezetimibe. Solid dispersion system adsorbed on a surface adsorbents could increase surface area of drug that contact with dissolution medium so that the dissolution rate of ezetimibe could increase. PEG 8000 was used as a hydrophilic carrier, and lactose monohydrate and Avicel PH 101 were used as adsorbents. The aim of this investigation was to improve the dissolution rate of the water-insoluble drug ezetimibe by preparing adsorbed solid dispersion ezetimibe-PEG 8000- (lactose monohydrate:Avicel PH 101=2:1) with ratio 1:1:8; 1:2:7; and 1:3:6. Adsorbed solid dispersion systems were made by melting method. Dissolution and powder X-ray diffraction (XRD) of adsorbed solid dispersion ezetimibe-PEG 8000-lactose monohydrate-Avicel PH 101, solid dispersion ezetimibe-PEG 8000 ratio 1:1; 1:2; and 1:3, and pure ezetimibe were evaluated. Differential Thermal Analysis (DTA) of solid dispersion ezetimibe-PEG 8000 were also evaluated. The result showed that the adsorbed solid dispersion of ezetimibe-PEG 8000-laktose monohydrate-Avicel PH 101 can improve dissolution rate of ezetimibe.
