Abstract

Virus pandemic H1N1-2009 has been spreading in the world in 2009 included Indonesia and caused 182.166 positive cases with 1.799 death. Recantly, resistance of oseltamivir was occurred. New antiviral drug are needed to prevent the resistance of oseltamivir. In this research, we tested Streptomyces sp. bacteria that have been isolated from compost soil in Rumah Kompos Bratang Surabaya (RKBS) and coded Streptomyces sp A, C, D, Ea, Ep, F, G and I. That isolated bacteria was fermentate d to pruduce secondary metabolite, and used the supernatant for antiviral activity test. In this study, an improved embryonated chicken egg model for evaluating antiviral activity against virus pandemic H1N1-2009 was developed. The virus that we used was A/Indonesia/UNAIR-367/2010. In the model, the virus was injected into the allantoic cavity and supernatant of Streptomyces RKBS was injected into the albumen of the egg. Supernatant of Streptomyces that inoculated at concentration 5% in PBS after toxicity test in embryonated chiken egg. The levels of virus pandemic H1N1-2009 in the allantoic fluid was titrated by the hemagglutination test after incubation for 72 h at 37°C and 12 h at 4 °C. The result, we found that at concentration 5% of supernatant of Stretreptomyces sp. D and Ep had titer HA value lower than viruses control. In conclusion, Streptomyces sp. D and Ep treatment showed antiviral activity against virus pandemic H1N1-2009 in this mode
