Abstract
Aerobic spore-forming bacteria such as Bacillus subtilis is a common contaminant found in microbiology laboratories, especially in the air. Bacillus subtilis contamination on MSA media can give false-positive results for Staphylococcus aureus because of the ability of Bacillus subtilis to ferment mannitol sugar. Sodium chloride with high concentration generally cause bacteria that are not resistant to sodium chloride will be stunted. This study was aimed to determine the growth of Staphylococcus aureus and Bacillus subtilis against some sodium chloride concentrations that were added to the nutrient agar to obtain certain concentrations of sodium chloride which only grown by Staphylococcus aureus but not overgrown by Bacillus subtilis. This study used a true experimental design. This study was using a treatment group (P1-P6) with the addition of different concentrations of sodium chloride in each group that was inoculated by Staphylococcus aureus and Bacillus subtilis, continued by incubating for 24, 48 and 72 hours at 37oC. The result showed the growth of both bacteria at 7.5% sodium chloride concentration at all incubation times. At the concentrations of sodium chloride 10%, 12.5%, 15%, 17.5%, and 20% only showed the growth of Staphylococcus aureus; meanwhile the growth of Bacillus subtilis was not seen on the media. Based on this study 10% to 20% sodium chloride concentrations could inhibit the growth of Bacillus subtilis while Staphylococcus aureus was not inhibited.
