Abstract

Background: Bone remodeling consists of two alternating processes, bone resorption and bone formation. bone graft can be used to repair bone damage. Broadly speaking there are several types of bone graft, one of which is xenograft. xenograft is a bone graft obtained from bovine bone donors. xenograft has various types, including Demineralized Freeze-Dried Bovine Bone Xenograft (DFDBBX), and Freeze-Dried Bovine Bone Xenograft (FDBBX). In the healing process, growth factor has an important role because of effective therapy. One of the bioactive factors needed from bone regeneration is fibroblast growth factor (FGF). FGFs are growth factors that regulate cell proliferation, migration, and differentiation in many organs including bone. The FGF family consists of many members, one of which is FGF-α. FGF-α is a powerful mitogen that functions in various types of cells in the body. Role in various stages of development and morphogenesis, as well as in angiogenesis and wound healing processes. Objective: To compare levels of fibriblast growth factor α in DFDBBX and FDBBX. Methods: In both samples, DFDBBX and FDBBX, protein extraction was carried out using the guanidine HCl method and centrifugation was performed. The supernatant is inserted into the plate and replicated 5 times for each sample to be tested for protein using ELISA Sandwich. Absorbance is read using ELISA reader with a wavelength of 450 nm. The absorbance value is then converted using dyna analysis software so that the FGF-α levels in DFDBBX and FDBBX (ng / mL) are obtained and data analysis is done using Mann Whitney Results: From the two samples, DFDBBX and FDBBX, the graft contains FGFα growth factor and more content in DFDBBX than in FDBBX.The average yield of fgf-α content in DFDBBX is 3,089 ng/ml FDBBX is 2,451 ng/ml. Teset result mann whitney p=3500 Conclusion: FGF-α levels in Demineralized Freeze-Dried Bovine Bone Xenograft are higher when compared to Freeze-Dried Bovine Bone Xenograft.
