Abstract

Ovalbumin is model of vaccine antigen which using enable increasing immunity. Ovalbumin-alginate microspheres could be develop to protect ovalbumin from pH and to formulate sustained release. Ovalbuminalginate microspheres were prepared by ionotropic gelation using aerosolization technique using alginate as polymer and calcium chloride as crosslinker. The objective of this study was to investigate effect of concentration of CaCl2 (cross-linker) and crosslinking time on characteristic of ovalbumin-alginate microspheres in term of morphology, particle size, protein loading, entrapment efficiency and yield. Results showed that ovalbumin-alginate microspheres had spherical shape and nearly smooth morphology. Increasing CaCl2 concentration from 0,5 M to 1,5 M reduced size of microspheres (from 18,06 μm to 12,54 μm). In addition, the particle size of microspheres decreased from (19,20 μm to 12,54 μm) by increasing crosslinking time (from 30 minutes to 2 hours). For Loading and entrapment efficiency, there were an increase by increasing CaCl2 concentration, however by increasing crosslinking time, the loading and entrapment decreased. Yield was so increased as CaCl2 and crosslinking time increased.
