Abstract

Chromatographic test of botanicals can be carried out for two purposes: identification and potency determination. The chromatogram profile may be used as refence standards which is compare with other raw materials. Alleged the differences of chromatogram profile will affect to the chemical constituents contained, that’s will affect to the activities. This experiment aimed to explain the chromatogram profile and antioxidant activity of methanol extract of black cumin seed based on the differences of particle size and variations of growth location. Samples was extracted by microwave assisted extraction (MAE). In this experiment TLC-Densitometry used for chemical constituents’ analysis and TLC-Bioautography used for antioxidant activity analysis. PCA (Principle Component Analysis) used for analysis the chemical constituents’, and antioxidant activity will be processed by multivariate analysis PLS (Partial Least Squares). Results of data processing PCA showed the sample grouping which means that variety affecting the chemical constituents of black cumin seed. PLS showed that variety affecting antioxidant activity, this can be seen from score-plot and bi-plot.
