Abstract

Colchicine is an alkaloid compound which can be isolated from Gloriosa superba plant. This plant is potential to be developed into herbal product and one of the most common herbal product is herbal drink. In this research, herbal drink was made from Gloriosa superba leaf infusion and its dose was adjusted according to colchicine dose. Determination of colchicine concentration in herbal drink is necessary to assure the dose was right. TLC-densitometry was used to determine colchicine concentration. Method validation is needed to assure if its analytic aspects were valid and the validation parametersmeasured in this research were selectivity, liniearity, precision, and accuracy. Chloroform:diethylamine (8:2) was chosen as mobile phase with maximum wavelength 354 nm. Liniearity was acquired from regresion function y= 4018,46x+6890,78 withcorrelation coefficient 0,9973 and Vxo = 3,68%. Meanwhile, its recovery was 91,26%. Colchicine concentration in leaf infusion was 0.1258% and colchicine weight in herbal drink was 5,228 x 10-3 mg/mL.
