Abstract

Validation method is a series of laboratory process used to assure that one analytical method have met all the validity requirements. Degree of validity can be measured by determining selectivity, linearity, accuracy, and precisy of the method. Analytical method that have met the requirements would produce a valid data, applied as well on determination of colchicine in granule. Colchicine is an anti – inflammatory agent indicated for the treatment of acute gout, and an antimitotic drug which is highly effective in preventing acute gout attacks. Colchicine as an antigout drug that have already formulated in tablet form now being formulated in dry powder form, therefore it can be pharmaceutically developed. Determination process of colchicine in granule is needed to be taken to fulfill the quality control step. The purpose of this study is to validate the UV Spectrophotometric method in terms of selectivity, linearity, precision and accuracy. The optimum condition was reached in wavelength 350,4 nm. In that condition, the equation of regression was y=0,0306x+0,0497, r = 0,9935 > r table for n=15, is 0,641 for p<0,01 and 0,514 for p< 0,05, Vxo = 5,12%, while the precision gave coefficient of variation 0,92% and the accuracy gave average % recovery of 84,79% with the coefficient of variation of 6,92%. The determination of colchicine in freeze dried extract and colchicine in dry powder was carried out in five replications. The result showed that the concentration of colchicine in freeze dried extract was 2,02% and the concentration of colchicine in powder was 0,4878 miligrams
