Abstract

This research produced ovalbumin-alginate microspheres using alginate and crosslinker BaCl2. The aim of this research was to investigate effect of concentration of alginate and BaCl2 on physical characteristics (protein loading, entrapment efficiency and yield). Microspheres were prepared by ionotropic gelation method aerosolization technique. The ratio of alginate and ovalbumin was 1:1. Concentration of alginate were 2,5 % and 3,5%, whereas BaCl2 concentration were 0,5 M and 0,75 M. The integrity of microspheres were also studied using SDS-PAGE. Results showed that ovalbumin-alginate miscrospheres had spherical and smooth particles with diameter decreased by increasing concentration of BaCl2. The size were F1 = 6,94 μm; F2 = 5,23 μm; F3 = 4,61 μm and F4 = 4,04 μm respectively. Protein loading were F1 = 66,86 % ± 10,36; F2 = 65,17 % ± 12,24; F3 = 61,34 % ± 4,16 and F4 = 53,59 % ± 2,70 while entrapment efficiency of F1 = 80,47 % ± 9,52; F2 = 81,81 % ± 10,77; F3 = 90,88 % ± 7,37 and F4 = 92,17 % ± 5,57. Yield of microspheres F1 = 60,63 % ± 3,06; F2 = 63,43 % ± 4,98; F3 = 62,93 % ± 5,96 and F4 = 73,82 % ± 10,57. Based on statistical analysis it means there were no significant differences in entrapment efficiency, protein loading and yield of all formulas. SDS-PAGE demonstrated that integrity of ovalbumin in the alginate microspheres was highly maintained by showing molecular weight of ovalbumin at 45 kDa. It can be concluded that by increasing polymer concentration and increasing crosslinking agent concentration, particle size decrease and there were no significant differences for ovalbumin content in microspheres, entrapment efficiency and yield of microspheres.
