Abstract

Mefenamic acid is an antiinflammatory drug. It has been classified as Biopharmaceutical Class II (BCS II) which has poor solubility in aqueous media and high permeability through membrane. Mefenamic acid-nicotinamide cocrystal with ratio of (1:1) and (1:2) has been studied by researcher as a method to improve mefenamic acid dissolution rate and stability. The aim of this study was to investigated phase diagram from physical interaction mefenamic acid-nicotinamide by differential thermal analysis (DTA), investigated physical characterization of co-crystallization product mefenamic acid-nicotinamide (1:1) and (1:2) by thermal DTA, powder X-ray diffraction and FTIR analysis spectroscopy and compare dissolution rate of co-crystallization product mefenamic acid-nicotinamide (1:1) and (1:2) with physical mixtures mefenamic acid-nicotinamide (1:1) and (1:2). Co-crystallization product mefenamic acid-nicotinamide (1:1) and (1:2) were made by melting method. The investigation phase diagram from physical interaction mefenamic acid-nicotinamide showed phase diagram type simple eutectic. The physical characterization result of co-crystallization product mefenamic acid-nicotinamide (1:1) and (1:2) was not formed cocrystal, but it showed the eutectic mixture. Dissolution rate showed significant increase in co-crystallization product mefenamic acid-nicotinamide (1:1) and (1:2) as eutectic mixture compare to the physical mixture and mefenamic substance.
