Abstract
This study investigated the effect of lyoprotectants on physical characteristics of ovalbumin-alginate microsphere during freeze-drying process. The parameters of physical characteristics of particle shape and size, entrapment efficiency, protein loading, and yield were studied. Microspheres were produced using aerosolization technique and were dried by freeze-drying method. The effect of type of lyoprotectant (lactose and maltodextrin) and concentration of lyoprotectant ( 5% and 10%) were investigated. Results showed that entrapment efficiency of F1, F2, F3, and F4 were 82,96 ± 5,98; 46,29 ± 6,81; 90,08 ± 6,16; and 54,15 ± 8,82 respectively. Protein loading of F1, F2, F3, and F4 were 46,11 ± 9,21; 28,07 ± 6,58; 48,32 ± 8,46; and 35,47 ± 1,60 where as yield of F1, F2, F3, and F4 were 60,35 ± 8,15; 41,41 ± 3,15; 61,51 ± 9,09; and 38,34 ± 7,61 respectively. Based on factorial design statistic analysis, variable concentrations indicated affected the entrapment efficiency, protein loading, and yield. Increasing concentration of both lactose and maltodextrin lyoprotectant decreased the entrapment efficiency, protein loading, and yield of microspheres. Moreover, there was no interaction between type of lyoprotectant and lyoprotectant concentrations that affected entrapment efficiency, protein loading, and yield. The microspheres size of F1, F2, F3, and F4 were 5,8157 μm; 5,9260 μm; 5,4645 μm; 5,0615 μm with spherical shape and smooth surface was formed.
