Abstract

Skin ageing becomes high problem during social interaction which can affect the self confidencies. Coenzym Q10 (CoQ10) is an antioxidant used for antiageing cosmetics which is naturally produced in the body; however, the level will decrease during ageing. Thus, requiring outside supplements to repair damaged skin conditions, especially in the skin dermis layer. A nanosized delivery system is urgently needed to increase the stability of CoQ10 was formulated as a protransfersomal gel mask. At first, CoQ10 was dissolved in oleic acid at the weight ratio of 1:56. Then, protransfersome loaded CoQ10 composed of L-α- Phosphatidylcholine and Tween 80 as the edge activator at molar ratio of 85:15, respectively, was prepared by a constant stirring at room temperature. Furthermore, the protransfersome loaded CoQ10 was dispersed in the emulgel base consisted of Tween 80 and Span 80. The physical characteristics included particle size, polydispersity index, entrapment efficiency, and stability during storage were evaluated. Moreover, the in vivo studies in male Balb/c mice were evaluated for skin penetration ability, antiageing activity, and irritability. The results showed that protransfersome loaded CoQ10 had homogeneous particles with a particle size of 201,5 ± 6,1 nm and a PDI value of 0,229 ± 0,047. In addition, it also had the Ph value 5,958 ± 0,046, which was acceptable for topical use. The protransfersome successfully encapsulated CoQ10 with entrapment efficiency of 45,64 ± 7,52 % and it was stable during storage. In the in situ hydration using normal saline, it could be confirmed that the protransfersome has changed into transfesomal vesicles. Then protransfersome loaded CoQ10 was being formulated as a protransfersome gel mask, it was obtained homogeneous particles with a particle size of 134,3 ± 4,8 nm and a PDI value of 0,291 ± 0,020. Protransfersomal mask had the pH value 5,958 ± 0,046 which also was acceptable for topical use. After 3 hours of topical administration, it has been shown that transfersome loaded CoQ10 succesfully reached the skin dermis layer. Moreover, the antiageing activity test showed that the CoQ10 protransfersome-loaded gel mask improved the collagen density by 74.65 ± 8.72% and the number of fibroblast up to 31.5 ± 9.48 cells per field of view. The 24-hour irritation study showed that there were no irritant signs observed during the study indicating that the CoQ10-loaded protransfersomal gel mask are safe to use. In conclusion, loading CoQ10 into protransfersomal gel mask successfully improved stability and antiageing efficacy providing its potential use as antiageing cosmetics.
