Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kasar polisakarida okra (Abelmoschus esculentus L.) terhadap kenaikan apoptosis dan nekrosis hepar mencit (Mus musculus) kanker terinduksi dietilnitrosamin. Hewan coba dibagi berdasarkan kelompok perlakuan, yaitu kelompok kontrol normal, kontrol negatif yang tidak diobati, kontrol positif yang diobati dengan doxorubicin, kelompok P1 yang diberi ekstrak kasar polisakarida okra (ORPE) 50 mg/kg BB, kelompok P2 yang diberi ORPE 100 mg/kg BB dan kelompok P3 yang diberi ORPE 200 mg/kg BB. Analisis sel hepar menggunakan uji flow cytometry menghasilkan data kuantitatif berupa persentase sel yang mengalami apoptosis awal, apoptosis akhir dan nekrosis dari masing-masing kelompok perlakuan. Data yang diperoleh memenuhi syarat uji positif analisis statistik. Hasil menunjukkan nilai persentase apoptosis awal tertinggi pada kelompok perlakuan ORPE terdapat pada kelompok P3 (12,17 ± 3,31)% dan terendah pada kelompok P1 (9,36 ± 1,91)%. Persetase apoptosis akhir tertinggi terdapat pada kelompok P2 (2,61 ± 0,50)% dan terendah pada kelompok P3 (1,30 ± 0,53)%. Sedangkan, persentase nekrosis tertinggi terdapat pada kelompok P3 (6,93 ± 0,86)% dan terendah pada kelompok P1 (4,48 ± 0,59)%. Dari hasil penelitian tersebut, disimpulkan bahwa hanya pemberian ORPE dosis 200 mg/kg yang mampu menaikkan persentase nekrosis, sementara dosis lain tidak mampu menaikkan persentase apoptosis awal maupun akhir dan nekrosis setara dengan obat doxorubicin.
