Abstract

p-methoxycinamic acid (PMCA) is a cinnamic acid derived compound obtained from hydrolysis process of ethyl p-methoxycinamic acid (EPMCA). PMCA has antinociceptive and analgesic activity but has low solubility in water (0.71 mg/mL at 25°C). Cocrystal formation is one of the methods to improve the solubility of PMCA. In this study, PMCA-caffeine cocrystal at 1:1 molar ratio made by microwave radiation method used water as medium reaction at concentration of 0%; 5%; and 10%w/w. The aim of this study was to determine the effect of addition water as medium reaction at concentration of 0%; 5%; and 10%w/w on physicochemical characteristics of cocrystal and to determine the result of APMS-caffeine cocrystal 10%w/w is better if compared to others. To confirm that cocrystals was characterized using Differential Scanning Calorimetry (DSC), Powder X-Ray Diffraction (PXRD), Fourier Transform Infrared (FTIR), and Scanning Electron Microscope (SEM). The result showed that the DSC thermogram showed decrease in the melting point of cocrystals compared to the melting points of constituent components with cocrystal 10%w/w has higher enthalpy than others. Difractogram of cocrystals showed a new diffraction peaks which were different from constituent components and physical mixture with difractogram of cocrystal 10%w/w has higher peak intensity than others. FTIR spectra of cocrystal showed the loss and shift of the absorption peak in O-H of APMS and C=O of caffeine. SEM micrograph showed that cocrystals had different surface morphology compared to their constituent components and physical mixture and cocrystal 10% w/w showed smaller size than others.
