Abstract
Sekar Putri Timur S. mutans, monoclonal antibodies, cytotoxicity, MIT assay, antibodies storage. Background. Based on Ministry of Health (2009), dental caries prevalence rate in Indonesia reached 89%. New alternative in caries prevention using monoclonal antibodies S. mutans 1 (c) 67 kDa that specific to cell surface protein 67 kDa of S. mutans. In 2001, Soerodjo et al produce monoclonal antibodies S. mutans 1 (c) 67 kDa with class tipe IgA, IgG1 , and IgG3 and have been storage in freezer with temperature -20 C from 2001 until 2010 at Pusvetma, Surabaya. As one of the dental health products, monoclonal antibodies S. mutans 1 (c) 67 kDa should have not toxic, and have properties biocompatibility. Long storage can affect change in the nature of the monoclonal antibodies. Purpose. The aim of this research is to determine the effect of cytotoxic fibroblast cells have been induced with monoclonal antibodies S. mutans 1 (c) 67 kDa class of IgA, IgG1, and IgG3 after storage. Method. Cytotoxicity test of monoclonal antibodies 1 (c) 67 kDa used MIT assay. Measurement of cytotoxicity was a optical density or absorbent. Value of absorbent in microplate showed the number of living cells in media culture. Result. 19A had lowest average optical density be compared with IgG] and IgGj. These results suggested that monoclonal antibodies S. mutans 1 (c) 67 kDa class type of IgA had lowest number of living cells be compared with IgGJ and IgGj. Conclusion. Monoclonal antibodies S. mutans 1 (c) 67 kDa class type oflgA has toxic properties be compared to IgGJ and IgGjafter storage.
