Abstract

The effect of some species of bacteria and sperrnlbacteria ratio on sperm quality has been studied. Four species of bacteria were used in this study : Staphylococcus epidermidis, Streptococcus /aecalis, Enterobacter aerogenes were obtained from semen culture of infertile men and E. coli was obtained from prostatic fluid culture from men with prostat and urinary system disturbances. Six semen samples fulfilling the WHO criteria (1992) were used in this study. After preparation by Percoll gradient-column method, sperm were inoculated in a microplate with Staphylococcus epidermidis, Streptococcus faecalis. Enterobacter aerogenes, E. coli under the spermlbacteria ratio I: 10 and 1: 1000. Sperm motility and aglutination was observed immediately, 3 and 6 hours after inoculation using an inverted microscope. At the second experiment, the detrimental influence of bacteria on sperm was prevented by adding penicillin. The design of this study was Randomized Control Group Extended Post Test Only Design. Collected data were analyzed by Anova same subject factorial. of which they would be examined futher by simple effect test • if the result was significant. The result of this research indicated that species of bacteria and sperm !bacteria ratio affecting the sperm motility grade a ( p < 0,05 ) and a+b (p < 0,05 ) after 3 and 6 hours of incubation. Conclution of this research indicated that species of bacteria and sperm !bacteria ratio affecting the sperm motility grade a and a+b in 3 and 6 hours after incubation. Staphylococcus epidermidis, Streptococcus faecalis, Enlerobacter aerogenes affecting the sperm motility in 6 hours after incubation, whereas E. coli affecting the sperm motility in 3 and 6 hours after incubation. The most detrimental effect on sperm motility was shown by E. cofi at the ratio ofspermibacteria 1:10 after 6 hours incubation. This detrimental effect was not prevented by the addition of penicillin. It appears that longer incubation period and higher sperm !bacteria ratio may cause lower sperm motility. Concerning sperm aglutination , Staphylococcus epidermidis and Streptococcus faecalis didn;n show any, whereas Enterobacter aerogenes at spennlbacteria ratio 1:10 cause spenn aglutination after 6 hours of incubation ,and E. coli cause spenn aglutination after 3 and 6 hours of incubation.
