Abstract

Retinoic acid is one form of lipid soluble vitamin A. It is excreted from the body slowly, so that an excessive dose will cause a toxic effect. In first trimester pregnant mothers, in particulary, it may result abnormalities in mild and severe congenital, and even fetal death. The objective of this study was to investigate the effect of the administration of retinoic acid in a dose of 60 mg/KgBW in pregnancy age of 10 days on reproductivity outcome and the abnormalities congenital of fetal external. This was a laboratory experimental study by reproductivity using randomized posttest only control group design. Experimental animals were female BALB/C strain mice (Alus musculus). A number of 16 pregnant mice from group treatment were induced by retinoic acid in a dose of 60 mg/Kg BW per oral, and 16 pregnant mice in control group were induced by sesame oil 1 mlihead. All experimental animals were induced in pregnancy age of 10 days, and then sacrificed within one day before delivery (pregnancy age of 18 days). Results revealed that mice induced by retinoic acid is shown of decreasing number of living fetus, increasing intrauterine deaths, which was including the number of died fetuses and resorbed embryo, was significantly different from 0,08 (p < 0.05) of those control group, while placenta implantation and fetal bodyweight was not significantly different (p > 0.05) from control group (p = 0.366, p = 0.711). Based on the data on living fetal percentage in control group, 80 living fetus (100%) had no abnormalities, which were different from group treatment, in which all 68 living fetus (58.2%) had abnormalities. In conclusion, retinoic acid in a dose of 60 mg/Kg BW that induced by pregnancy mice age of 10 days will decrease the reproductivity the number of living fetus, increasing dead fetus, and resorbtion. Placenta implantation and fetal body weight is not affected, while external abnormalities occurs in all living fetus.
