Abstract

The development of early pregnancy diagnosis method in mammals can be undertaken by observing the specific substance present within mother's blood, the Pregnancy Specific Protein B (PSPB). This study was conducted through the following stages: Diagnosis of dairy cattle pregnancy with PSPB microtitre strip on day 21 post-artificial insemination and Pregnancy test by determination of serum progesterone level was carried out to confirm the pregnancy diagnostic by PSPB microtitre strip. PSPB protein level of dairy cattle blood serum obtained in days 21 of post-artificial insemination was 320.375-557.875 ug/ml. The result of dairy cattle blood serum progesterone on day 21 post- artificial insemination revealed that 32 dairy cattle had progesterone level of > 1 ng/ml (1.18-5.96 ng/ml) and 18 dairy cattle had progesterone level of <1 ng/ml (0.07-0.93 ng/ml). The result of PSPB microtiter strip with blood serum progesterone level test revealed sensitivity, specificity, accuracy, positive predictive value, and negative predictive value of 94.29 %, 80%, 91.67 %, 87.71% and 90%. Based on the results of the study and data analysis, a general conclusion could be drawn as follows: anti-PSPB can be employed to diagnose pregnancy in dairy cattle by means of PSPB microtiter strip method.
