Abstract

Laserpunctureisoneoftheapplicativetechnologiesusedmainlyinanimaland fish reproductions. Laserpuncturetechnologyhasbeenusedtoimprovegonadalmaturityandspermqualityin fish rapidly. This studyaimedtodeterminetheeffectsofdifferentlaserpuncturedosesongonadalmaturityand sperm qualityofmalestripedcatfish. Malesstripedcatfish (800e900g/fish bodyweight)andIgonadal maturitystagewereused.Semi-conductorsoftlaserwasusedwithdosesof0.2-,0.4-,and0.5-J,while the negativecontrol(withoutthelaserpunctureandtheovaprim™) andonlytheovaprim™ wereused as acomparisontreatment,respectively.Thesoft-laserwastreatedonreproductiveacupointeveryweek for fourweeks,whiletheovaprim™ wasadministeredbyintramuscularinjectionatdoseof0.2mL/kg fish in final rearingperiod.Fishwasrearedinhapaatthecontrolledpond.Fishwasfedwithacom- mercial feedcontaining32%crudeprotein.Gonadalmaturity,gonadosomaticindex(GSI),hepatosomatic index(HSI),andspermqualityofmalestripedcatfish weremeasuredinthe final rearingperiod.The results showedthatthelaserpunctureonthereproductiveacupointhadahighlysignificanteffect (P < 0.01)onthegonadalmaturity,GSI,HSI,andspermqualityofmalestripedcatfish. Intermsofthe gonadal maturity,laserpuncturedosestreatmentof0.4and0.5-JgavethemostmatureIVstage.While the highestlevelsofGSIandHSIwerefoundin0.5-Joflaserpuncturedose,whichwas2.17%and1.54%, respectively.Inaddition,thebestspermqualitieswereobservedin0.5-Joflaserpuncturedose,which were81.75%motility,82.75%viability,and7.0 � 109 cell/mLconcentration.Theseresultssuggestthatthe laserpuncturecanaccelerateagonadalmaturityandimprovespermqualityinmalestripedcatfish.
