Abstract
The aim of this study to observe the effectiveness of Haemopoitic Stem Cell in pregnant mice exposed to plumbum, and the observed cases are the numbers of necrosis cells and caspase-3 expression in the placenta. Haemopoitic Stem Cells was expected to able to reduce the number of necrosis cells and was abled to reduce the expression of caspase- 3. This study used a randomized design, and the required samples in this study were 40 female mice and 40 male I mice (Mus musculus). Each treatment utilized 10 male mice and 10 female mice with details as follow, (1) first treatment group (Tl): pregnant mice were given a dose of plumbum 1112 LD50 (54.42 mg I ml) during gestation days 9-15 and on day 16 distilled water was given orally. (2) second treatment group (T2): pregnant mice were given a dose of plumbum 1116 LD50 (40.75 mg I ml) during gestation days 9-15 and on day 16 distilled water was given orally. (3) Third treatment group (T3): Plumbum pregnant mice were given a dose of 1112 LD50 (54.42 mg I ml) during gestation days 9-15 and on day 9 Hemopoetic Stem Cell lx107 was given intravenously. (4) fourth treatment group (T4): pregnant mice were given a dose of Plumbum 1116 LD50 (40.75 mg I ml) during gestation days 9-15 and on day 9 Hemopoetic Stem Cell lxl07 was given intravenously. The obtained data were analyzed by non-parametric and parametric analysis. Nonparametric analysis used the Kruskal-Wallis test, if the data are significantly different, they will be followed by Mann Whitney Test. Analytical analysis used univariate analysis, if there was an interaction between the factors studied, it was then followed by one-way analysis of variance (ANOV A) I one-way ANOV A. The analysis was performed by assuming that the tested population was normally distributed, homogenous, and the samples were not related to each other. The results of the expression of caspase-3 between treatments are significant, especially the fourth treatment group (T4). Of all the treatments (Tl, T2, T3) when ompared to treatment P4 shows a significantly different result. T4 was treated by giving Hemopoetic Stem Cells on day 9 which coincided with the first day of plumbum exposure. Intended stem cells function properly because the homing process is good enough for stem cells.
