Abstract

The main purpose of this study is to evaluation the relation between amount of capacitation, uptake Ca2+, and cytochrome c expression cytosolic in Peranakan Ettawa frozen semen and correlation of viability and motility with difference thawing duration. This study used 22 mini straw frozen semen goats with 4 difference treatments, semen that was thawing on 27°C for 10 second as control (PO), Pl semen was thawed at 37°C for 10 second, P2 was thawed at 37°C for 20 second, and P3 was thawed at 37°C for 30 second. Capacitation was detected by using chlortetracycline coloring (CTC). The positive result showed fluorescent yellow at two-thirds at sperm head region. Uptake Ca2 + was detected by using Fluo-3 coloring than observed use Confocal Laser Scanning Microscope (CLSM) and the results are expressed in intensity / μm2 • Cytochrome c cytosolic expression was detected by using immunohistochemistry coloring with Ultra Vision Detection System. The positive result showed tan brown at sperm neck. Based on data analyzed by using Multivariate Analysis of Variant for all treatment and test followed by Hotteling 's Trace 5%, the result indicated that there is significant difference (p<0,05) between motility, viability, capacitation, uptake Ca2+in sperm post-thawing of Peranakan Ettawa goat with difference duration. Thawed at 27°C for IO seond showed lowest percentage on motility, viability, capacitation, uptake Ca2 + and cytochrome c cytosolic expression sperm.
