Abstract

This study was aimed to detennine the potential dose of sambiloto extract (Andrographis paniculata) as an immunostimulant in PBMC cachama (Colossoma macropomum) on the expression of IFN-y and TNF-a by in vitro. Blood from a 8 months cachama with 600 grams weight was used for samples. 10 ml of a cachama blood was taken and grouped into 30 well to incubated for two days with doses 5 mg I ml, I 0 ug I ml, 15 ug I ml, 20 ug I ml, 25 ug I ml sambiloto extracts and a control treatment which was given the sambiloto extract solvent (DMSO) without any sambiloto extract as a comparison. Results in PBMC cultures showed that sambiloto extracts could induce the expression of various cytokines and cells because it had an active ingredient diterpenoid and flavonoids which was indicated by a cell growth in culture media. In the chemical immunocytochemistry test, sambiloto extract (Andrographis paniculata) proved the capability of being immunostimulatory cachama PBMC which could express IFN-y and TNF-a (Colossoma macropomum). The average percentage of IFN-y and TNF-a were expression in PBMC cells for Pl group (dose 5 mg I ml) showed the highest results are 97.9% and it was 96.4% significantly different (p <0.05) when compared with the treatment group PO (control treatment), P3 (dose of 15 mg I ml), P4 (dose of 20 mg I ml) and P5 (dose of 25 mg I ml), but were not significant difference (p> 0.05) when compared with P2 (dose of 25 ug I ml). A.paniculata extracts could produce IFN-y from NK cells, CD8 cells, CD4 Thi cells and also activate macrophages to release IL-12 and TNF-a. As a conclusion from this study can be avowed that the sambiloto extract (Andrographis paniculata) can take the role as an immunostimulant in PBMC cachama ( Colossoma macropomum) which is indicated by the presence of IFN-y expression lymphocytes and TNF-u expression monocytes by in vitro at the optimum dose 5 mg I ml. In addition, it needs a further research by in vivo and a test with a variety of diseases.
