Abstract

"Crassostrea iredalei is an oyster species with the highest commercial value compared to other oyster genera in Malaysia. The challenge that Malaysian C. iredalei oyster farmers experience is misidentification due to the high plasticity of oysters. Molecular techniques are a viable solution in classifying oysters to improve understanding of species diversity and taxonomy. DNA extraction is the initial stage of the genetic analysis method of C. iredalei oysters. In the process, DNA extraction can support the process of genetic manipulation, as well as the purification and restoration of DNA that is more efficient, short, and with high-quality results. Institute of Tropical Aquaculture and Fisheries (AKUATROP) is a research institute of Universiti Malaysia Terengganu that is actively engaged in the research and development of aquaculture including in the field of molecular genetics. Therefore, Field Work Practices were carried out at AKUATROP with the aim of learning the C. iredalei DNA extraction process in Malaysia, from October 17 2022 to February 28 2023. The method used in this field work practice is a theoretical method with data collection including primary data and secondary data. DNA extraction is intended to determine the concentration and quantity of nucleic acids for further genetic analysis procedures. The DNA extraction process consists of tissue isolation, membrane lysing, extraction, purification, and precipitation. In order to analyze the quality of DNA samples, the purity and quantity of DNA are also measured using a NanoPhotometer machine and gel electrophoresis. Studying DNA extraction techniques is important because it is applicable in fisheries, such as the identification of trait decline patterns, species identification, genetic population analysis, genetic diversity analysis, and conservation."
