Abstract

Non-Alcoholic Fatty Liver Disease (NAFLD) is caused by the accumulation of fat in the liver, followed by inflammation, and leads to fibrosis. Liver macrophages have a major role in inflammation. This study aimed to analyze the role of resveratrol on macrophage polarization. This study used male-ddY-mice which were divided into 4 groups including Normal Chow; High Fat Diet (HFD); HFD+Resveratrol 15 mg/KgBB; and HFD+Resveratrol 30 mg/KgBB. All food was given ad libitum. Resveratrol was administered intraperitoneally from days 15-28. Based on the results, it is known that the model has developed into an early NAFLD stage, characterized by lipid accumulation and mild inflammation. Triglyceride levels in the serum of the HFD group (±233 mg/dL) increased significantly in the normal group (±99 mg/dL). Resveratrol suppresses serum triglyceride levels (±73 mg/dL), but it was not significant in both doses. The accumulation of fat in the liver was shown by the macroscopic appearance of the liver of the HFD group which was pale. The abnormalities in hepatocyte morphology of HFD were confirmed by microscopic appearance. Resveratrol is known to improve the condition of NAFLD both macroscopically and microscopically. RT-PCR data showed that there was a tendency for macrophage involvement in the inflammatory process, but there was no polarization between macrophages 1 and 2. Both doses of Resveratrol had no impact on macrophage polarization. Body weight was measured daily, HFD induction caused weight loss of 35% from baseline, and the normal group had a 28% increase from baseline. However, Resveratrol did not affect body weight.
